Usage of the EMBL-incubation box (installed on SP5 microscope)
- If using CO2 open the valves on the CO2 flask, final output pressure should be between 1 and 1.5 bar (third manometer).
- Pressure can be fine tuned by turning the middle black screw between the manometers.
- The main switch (red, on the wall) of the control unit needs to be ON.
- The second red switch (wall) controls the neon lamp inside the box.
- All other functions can be set with a turnable and pressable button on the small control box.
- Turn the button in any direction.
- 'Answer' the questions on the display by yes/no, confirm by pushing the button.
- Set the temperature and CO2 by turning the button followed by confirmation (press button).
- After completing setup start incubation box by selecting 'Start', yes (press button)
- The microscope will need one hour to stabilize at 37°C and roughly 15 min to reach eg. 5% CO2
- Therefore, leave incubation box on, if the next user requires it, too.
- If using CO2: during sample change press the CO2-OFF button (green on control unit) in order to temporarily block CO2 supply, don't forget to unblock it!
- Many cell lines can be kept without CO2 during microscopy in special media such as Leibovitz L15, CO2-independent medium and Hepes buffered formulations.
- Whenever possible, use phenol-red free media since phenol red is highly autofluorescent.
